Speechless day..

tired all day long, sampling as usual...withdrawing samples, BOD, ph...and with additional things happen, so i have additional work too..n im speechless..

Karaoke time

yeay! feels so refreshing...We went for a karaoke session for 2 hours at Music Box, Plaza Perangsang Shah Alam...Parked our car at the basement of Plaza Alam Central and just walk to the Quality Hotel..find the lift and press the button of 24! here we go, but opssss! another stairs ya! its level 25...so I cant bring mother along next time..hehe she wont even bother to follow! haha

I was so stressed out yesterday coz I couldnt finish up my abstract...I dont think I wanna go the Miri Conference..im not ready....ask for Zaki's opinion, he told me to let Prof know that Im not ready...Stress again this morning, so I try to email Prof...explaining so many things to him..

~I'll amend the abstract but i dont think I can send it to the CUTSE Miri, since i haven't got any results yet..let me use the results for another conference paper or journals..I'm not ready yet..so sorry, find it very hard to tell you..

I need more time to compile my results for journals too, at least few more runs for the metal sulphide precipitation, because i couldn't yet to prove that the rest of metals that was not reacted with sulphide was actually undergoing absorption process ..hope prof okay with it..n in August, Im going to proceed with molecular techniques, so ill stop my experiment on BOD and also metal sulphide precipitation.~

n he replied..."It's okay, keep up the hard work"...

reading that would just make me smile, gambattee..will word even harder! Thanks Prof!

we went to Carefour Sect 24 last friday, purposely go to buy keel for my brother..he is comin on saturday morning..mother, normal lah, love him so much..the only thing that my brother eat is chicken...weird rite? only keel...huh...unlike me, hehehe

after finished shopping, we went to this restaurant at the end of the mall...

sis ordered black pepper chicken chop which is okay but mine is terrible, chicken chop with brown sauce, feels like eating chicken kurma..what a weird taste...a small restaurant with a bill of service charges for almost 10%. can u imagine that?

nowadays, we avoid going to restaurant that charges us, just to boycott!

i just dont reccomend this restaurant..not that great, drinks were not so nice too, just dont try it but it the end, it all depends on u! ...if u wanna try, just go on..

• went to environment laboratory, level 3..(BOD, withdraw samples and measuring pH)
• bought some lauk for shedan, he wanted tempe at medan selera section 7 shah alam
• went back home, lunch and wait for husna to arrive..

BOD-some basic explanation

BOD (Biochemical oxygen demand) is a measure of the amount of oxygen that bacteria will consume while decomposing organic matter under aerobic conditions.

COD (Chemical oxygen demand) is a measure of the total quantity of oxygen required to oxidize all organic material into carbon dioxide and water.

*COD values are always higher than BOD values

DO meter, step by step..

This is only for YSI 5000 model for DO meter

Switch on, wait for 15 minutes (until stable value appear) then only press CALIBRATE..follow the procedure as in pictures below:

press calibrate, leave for few minutes until stable condition achieved

press autocalibrate, wait until it appears as "DO calibration saved"

press mode to go back to original screen

now only measurement can be taken after calibration has been done and saved!!

Make sure probe is always in good condition...maintaining it is very important..

Probe should be cleaned, color as the pic above

• membrane should be changed frequently if usage is high
• if possible, look at the KCl as well, to prevent from any damage on the membrane, add KCl if its not enough, must prevent from any bubble when adding KCl..be careful
• probe should be change at least once a year
• must wipe membrane with soft tissue after rinsing it with distilled water
• extra precaution must be taken to prevent from any damage to membrane, its expensive. cost about rm400/membrane
• price for probe, rm4000
• price for meter, rm10000
• so take extra careful when using DO meter..its so expensive!

Step by step in preparation of BOD using DO probe

Step by step:

1. Preparation of Dilution Water
2. Preparation of BOD bottles
3. Read the dissolved oxygen content in the five BOD bottles using a dissolved oxygen probe. Ensure that no water is lost from the BOD bottles. Replace the stoppers without trapping any air.
4. Place the BOD bottles in the incubator. To prevent oxygen from leaking into the bottles during the incubation period, fill the portion of the bottle around the stopper with water using a squeeze bottle. It is the responsibility of the laboratory group to ensure that the water seal on the BOD bottles is maintained by checking each day and replenishing as required using a squeeze bottle of distilled water.
5. Following 5 days, remove the BOD bottles from the incubator and determine the dissolved oxygen concentration of each.
6. Compute the BOD using the following formula

BOD mgL= [(Initial DO-Final DO) x 300mL] / mL sample

Im currently waiting for my primers to arrive, the supplier will send it to me at any time now...

Ive ordered 14 primers, its for the fingerprinting of Sulphate reducing bacteria (SRB) in my biofilm reactor! Im planning to proceed with the DNA thing during the fasting month, so i dont have to take wastewater sample during that month..Pity those Bangladesh people, coz they are the one who help me to sample and carry the 30L of wastewater.

Oh by the way, Ive received my chemicals yesterday, manganese (II) nitrate tetrahydrate and zinc (II) nitrate hexahydrate..I can just proceed coz I already have the 3 metals that I want!

ICP-LESSON 1

things that u must make sure before running ICP,

1. check gas
2. tore samples
3. check tubing

To warm up ICP:

1. Switch on plug of ICP, PC, Chiller
2. Check gas, open the gas tank, both on the gas tank & on the wall to green color.
3. Check air compressor (push the red button to the back, open the blue screw on the right and 1 more screw on the bottom. Shake that air compressor, just to make sure, water flows out coz condensation occurs.
4. Re-tighten the screw back and pull the red button to the front.
5. On ICP generator button, its on the right side.
6. Open the winlab 32
7. Click F11
8. Lock the yellow and white tube
9. To wet tubing, click on Plasma control
10. On pump
11. After no bubble seen, off the pump
12. On plasma, wait to 5 mins
13. stabilize, minimize window for plasma control
14. Click Auto, cancel -Print log during analyses
15. Result data set name
16. open, re-name; name and description
17. click analyze
18. check back with the Location and Samples ID type previously
19. If OK, click analyze ALL
20. go to Spectra to check for calibration

sis was with me today, helping...(helping? i pay her okay!) but not much la, just to help with few things...

as usual, sampling, transport 30L++ wastewater to Lab level 2, withdraw old wastewater, replace with new one, purge almost 30mins with nitrogen gas, withdraw few sets of samples and thats it! & dont forget to cover it up with aluminium foil!

• hahaha, running samples for both compounds-sulphate using IC & metals using ICP..
• Ive warmed up both instrument, IC requires at least 1hour, ICP requires 3 hours..
• Ive prepared standards, acids, blank..so now, time for me to rest while waiting for the instrument...

• Now i have some time to check emails and write down here in my blog..

• usually, the most busiest time of the day is around 8-11am. the rest from 11-5pm, im a bit free, i can use that time to check blog, write up, check email again and so on..

Haloo, morning people! yesterday, my uncle came n visit us...the best part is, going to meet HUSNA! hehehe she is so chUBBY!...weight about 17kg now! wow, heavy.....I cant even lift her up..huhu Ive attached her pics as below:

FYI, its frozen yogurt not ice cream as others have mentioned previously..I dont know y, it is called ice cream while its not, i love this but its just that i think this branch is just not to what we expected...Im going to try this again at another outlet...cost us about RM23 for 2 cup filled with various flavors and toppings! enjoy yours too ya!

Finally, I got the answers already!!

I didnt use the method of calculating as suggested by the company, I just use the normal maths calculation..

1. calculating the total surface area of bioblock (14)
2. calculating the surface area of reactor (cylinder)
3. Total surface area of biofilm= (1)+(2)....TADA!

the unit for biological sulphide produced per biofilm surface area would be in mg S/L.m2
