Things that u must have, or need to consider to buy before starting on molecular work.

- EDTA biotechnology grade-Bioshop Disodium salt, dihydrate 1kg/500g
- Schott bottle, 50ml, 500ml, 1L
- Beaker 500ml
- Vol flask 100ml

- Magenetic bar
- Tips rack
- Falcon tube rack for 50ml and 15ml
- Gloves

Tips before starting on molecular work
Ive sent this notes to friends who r going to do on molecular work, thought of sharing this too with u guys..

Hai,
before beginning ur lab on molecular work.here are some tips that could help u guys. Pls take note.
- U have to check what are the bacteria u want, intend to determine.
- Pls check what are the primers from the literature review. Is it general primers or specific primers? It depends on ur objective of work. It could be different from one journal to another, u may try few generals and few specific.
- Once uve checked the primers and uve already decided which primers that u want to use, pls get their sequence. Usually u have to look at the old journals paper, then only u can get the sequence. Dont re-type, pls copy and paste to the word. Then, u can send the list of primers with sequence to the supplier.
- Check on the overall method- method of extraction, protocol, storage condition, tips to prevent contamination.
- Read and check on- DNA extraction-how to extract, chemicals needed, temperature storage, consumable items.
- Read and check on - PCR, real time or normal PCR, how to prepare samples, how to dilute primers, how to store samples, chemicals needed, protocol of running samples, annealing temperature for ur primers
- Read and check on- Gel electrophoresis, how to prepare, what are the ingredients, chemicals required, method, storing, calculation of preparation of gel. how long would it run, what is the voltage required.
- Read and check on-DGGE, chemicals required, protocol, primers.
- Read and check on-Sequencing, phylogenetic tree, which company and etc.
- Make sure all the chemicals and consumable item required are available before u start the experiment.
- Make sure u know the calculation for each chemicals required to prepare.
- There are lots of tips and guidance on molecular work in the internet, u can have a look at it first to get the overall view of what are the processes that u have to go through. will be very2 difficult at 1st. Good Luck.
